Are you closer to York University or Union Station?

From Toronto Union, you can take the train towards Hamilton, get off at Aldershot or Burlington (depending if it's rush hour or not) then take the 15 (from Aldershot) or 15A. They'll usually announce on the PA system when you reach Burlington/Aldershot which GO buses are available at that stop.
That's the only route that I know of xD

From the Union GO station, take the Hamilton Express bus (I believe it's #16... you can check on the schedule board) straight to Hamilton. It will drop you off at Main and Longwood (beside Columbia college, where the highway exits on Main St W.).
If you're closer to York University, take the #47 bus to Hamilton at the university bus loop (where York Lanes is) and it will drop you off right on campus.

That is definitely the easiest route. Get to Union for the train that leaves at xx:43, and ride it until Aldershot. From there, follow the crowd of people to the bus area and take the 15 to McMaster. It only runs to Mac, not from.
EDIT: On Sundays, for clarification. Forgot to include that.

Are you talking about the 15? Because the 15 goes from Mac as well.

On Sundays, no; only the 18 from the GO Centre does.

I thought I had included "on Sundays", but I guess I didn't.

No. At Union, you buy a ticket to McMaster. Then when you get to the bus, show the driver that same ticket.
