That's weird... It's open for me, and it has been for a while now. Could it be because I'm registered in it?
Although I checked another full class that I'm registered in (Biology 2B03, in this case) and it says it's full. So I'm not sure why it says 2L03 is open for me.

So this is what I discovered last week.
If you email or go to the department office, you can ask them if they have space in the course. In most cases, they do and you just request a place in the class.
A lot of trouble and well it sucks having to do this for every core class.

It probably won't work for a course like this. 40 spots and LOTS more biology students who want it. It's not a typical lecture course so that's probably why.
